Intended Uses: This PS2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Goat PS2. This ELISA kit for research use only!

Principle of the Assay: PS2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PS2 antibody and an PS2-HRP conjugate. The assay sample and buffer are incubated together with PS2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PS2 concentration since PS2 from samples and PS2-HRP conjugate compete for the anti-PS2 antibody binding site. Since the number of sites is limited, as more sites are occupied by PS2 from the sample, fewer sites are left to bind PS2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PS2 concentration in each sample is interpolated from this standard curve.

NCBI Description

Alzheimer's disease (AD) patients with an inherited form of the disease carry mutations in the presenilin proteins (PSEN1 or PSEN2) or the amyloid precursor protein (APP). These disease-linked mutations result in increased production of the longer form of amyloid-beta (main component of amyloid deposits found in AD brains). Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor such that, they either directly regulate gamma-secretase activity, or themselves act are protease enzymes. Two alternatively spliced transcript variants encoding different isoforms of PSEN2 have been identified. [provided by RefSeq, Jul 2008]
