Chicken Prostaglandin E Receptor 3 ELISA Kit
Principle of the Assay: EP3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-EP3 antibody and an EP3-HRP conjugate. The assay sample and buffer are incubated together with EP3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the EP3 concentration since EP3 from samples and EP3-HRP conjugate compete for the anti-EP3 antibody binding site. Since the number of sites is limited, as more sites are occupied by EP3 from the sample, fewer sites are left to bind EP3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The EP3 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the G-protein coupled receptor family. This protein is one of four receptors identified for prostaglandin E2 (PGE2). This receptor may have many biological functions, which involve digestion, nervous system, kidney reabsorption, and uterine contraction activities. Studies of the mouse counterpart suggest that this receptor may also mediate adrenocorticotropic hormone response as well as fever generation in response to exogenous and endogenous stimuli. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Aug 2009]

The activity of this receptor can couple to both the inhibition of adenylate cyclase mediated by G-I proteins, and to an elevation of intracellular calcium.
