Human Hepcidin Antimicrobial Peptide ELISA Kit
Principle of the Assay: HAMP ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-HAMP antibody and an HAMP-HRP conjugate. The assay sample and buffer are incubated together with HAMP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HAMP concentration since HAMP from samples and HAMP-HRP conjugate compete for the anti-HAMP antibody binding site. Since the number of sites is limited, as more sites are occupied by HAMP from the sample, fewer sites are left to bind HAMP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HAMP concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
The product encoded by this gene is involved in the maintenance of iron homeostasis, and it is necessary for the regulation of iron storage in macrophages, and for intestinal iron absorption. The preproprotein is post-translationally cleaved into mature peptides of 20, 22 and 25 amino acids, and these active peptides are rich in cysteines, which form intramolecular bonds that stabilize their beta-sheet structures. These peptides exhibit antimicrobial activity. Mutations in this gene cause hemochromatosis type 2B, also known as juvenile hemochromatosis, a disease caused by severe iron overload that results in cardiomyopathy, cirrhosis, and endocrine failure. Uniprot Description
HAMP: Seems to act as a signaling molecule involved in the maintenance of iron homeostasis. Seems to be required in conjunction with HFE to regulate both intestinal iron absorption and iron storage in macrophages. Defects in HAMP are the cause of hemochromatosis type 2B (HFE2B); also known as juvenile hemochromatosis (JH). HFE2B is a disorder of iron metabolism with excess deposition of iron in the tissues, bronze skin pigmentation, hepatic cirrhosis, arthropathy and diabetes. The most common symptoms of hemochromatosis type 2 at presentation are hypogonadism and cardiomyopathy. Belongs to the hepcidin family.

Product Notes
The Human HAMP hamp (Catalog #AAA746152) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA746152 ELISA Kit recognizes Human HAMP. It is sometimes possible for the material contained within the vial of "Hepcidin Antimicrobial Peptide, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
