Principle of the Assay: MIS/AMH ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MIS/AMH antibody and an MIS/AMH-HRP conjugate. The assay sample and buffer are incubated together with MIS/AMH-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MIS/AMH concentration since MIS/AMH from samples and MIS/AMH-HRP conjugate compete for the anti-MIS/AMH antibody binding site. Since the number of sites is limited, as more sites are occupied by MIS/AMH from the sample, fewer sites are left to bind MIS/AMH-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MIS/AMH concentration in each sample is interpolated from this standard curve.

NCBI Description

Anti-Mullerian hormone is a member of the transforming growth factor-beta gene family which mediates male sexual differentiation. Anti-Mullerian hormone causes the regression of Mullerian ducts which would otherwise differentiate into the uterus and fallopian tubes. Some mutations in the anti-Mullerian hormone result in persistent Mullerian duct syndrome. [provided by RefSeq, Jul 2008]

AMH: This glycoprotein, produced by the Sertoli cells of the testis, causes regression of the Muellerian duct. It is also able to inhibit the growth of tumors derived from tissues of Muellerian duct origin. Defects in AMH are the cause of persistent Muellerian duct syndrome type 1 (PMDS1). PMDS1 is a form of male pseudohermaphroditism characterized by a failure of Muellerian duct regression in otherwise normal males. Belongs to the TGF-beta family.
