Principle of the Assay: NP-Y ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NP-Y antibody and an NP-Y-HRP conjugate. The assay sample and buffer are incubated together with NP-Y-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NP-Y concentration since NP-Y from samples and NP-Y-HRP conjugate compete for the anti-NP-Y antibody binding site. Since the number of sites is limited, as more sites are occupied by NP-Y from the sample, fewer sites are left to bind NP-Y-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NP-Y concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a neuropeptide that is widely expressed in the central nervous system and influences many physiological processes, including cortical excitability, stress response, food intake, circadian rhythms, and cardiovascular function. The neuropeptide functions through G protein-coupled receptors to inhibit adenylyl cyclase, activate mitogen-activated protein kinase (MAPK), regulate intracellular calcium levels, and activate potassium channels. A polymorphism in this gene resulting in a change of leucine 7 to proline in the signal peptide is associated with elevated cholesterol levels, higher alcohol consumption, and may be a risk factor for various metabolic and cardiovascular diseases.
