Principle of the Assay: PYGM ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PYGM antibody and an PYGM-HRP conjugate. The assay sample and buffer are incubated together with PYGM-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PYGM concentration since PYGM from samples and PYGM-HRP conjugate compete for the anti-PYGM antibody binding site. Since the number of sites is limited, as more sites are occupied by PYGM from the sample, fewer sites are left to bind PYGM-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PYGM concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a muscle enzyme involved in glycogenolysis. Highly similar enzymes encoded by different genes are found in liver and brain. Mutations in this gene are associated with McArdle disease (myophosphorylase deficiency), a glycogen storage disease of muscle. Alternative splicing results in multiple transcript variants.[provided by RefSeq, Sep 2009]

PYGM: an important allosteric enzyme in carbohydrate metabolism. Enzymes from different sources differ in their regulatory mechanisms and in their natural substrates. However, all known phosphorylases share catalytic and structural properties. Dimers associate into a tetramer to form the enzymatically active phosphorylase A. Phosphorylation of Ser-14 converts phosphorylase B (unphosphorylated) to phosphorylase A.
