Principle of the assay: CEBP? ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CEBP? antibody and an CEBP?-HRP conjugate. The assay sample and buffer are incubated together with CEBP?-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CEBP? concentration since CEBP? from samples and CEBP?-HRP conjugate compete for the anti-CEBP? antibody binding site. Since the number of sites is limited, as more sites are occupied by CEBP? from the sample, fewer sites are left to bind CEBP?-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CEBP? concentration in each sample is interpolated from this standard curve.

NCBI Description

The C/EBP family of transcription factors regulates viral and cellular CCAAT/enhancer element-mediated transcription. C/EBP proteins contain the bZIP region, which is characterized by two motifs in the C-terminal half of the protein: a basic region involved in DNA binding and a leucine zipper motif involved in dimerization. The C/EBP family consist of several related proteins, C/EBP alpha, C/EBP beta, C/EBP gamma, and C/EBP delta, that form homodimers and that form heterodimers with each other. CCAAT/enhancer binding protein gamma may cooperate with Fos to bind PRE-I enhancer elements. Two transcript variants encoding the same protein have been found for this gene.
