Specificity
This assay has high sensitivity and excellent specificity for detection of CAMKK2. No significant cross-reactivity or interference between CAMKK2 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CAMKK2 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This CAMKK2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse CAMKK2. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CAMKK2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CAMKK2 antibody and an CAMKK2-HRP conjugate. The assay sample and buffer are incubated together with CAMKK2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CAMKK2 concentration since CAMKK2 from samples and CAMKK2-HRP conjugate compete for the anti-CAMKK2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CAMKK2 from the sample, fewer sites are left to bind CAMKK2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CAMKK2 concentration in each sample is interpolated from this standard curve.

NCBI Description

The product of this gene belongs to the Serine/Threonine protein kinase family, and to the Ca(2+)/calmodulin-dependent protein kinase subfamily. The major isoform of this gene plays a role in the calcium/calmodulin-dependent (CaM) kinase cascade by phosphorylating the downstream kinases CaMK1 and CaMK4. Protein products of this gene also phosphorylate AMP-activated protein kinase (AMPK). This gene has its strongest expression in the brain and influences signalling cascades involved with learning and memory, neuronal differentiation and migration, neurite outgrowth, and synapse formation. Alternative splicing results in multiple transcript variants encoding distinct isoforms. The identified isoforms differ in their ability to undergo autophosphorylation and to phosphorylate downstream kinases. [provided by RefSeq, Jul 2012]

Uniprot Description

CAMKK2: Calcium/calmodulin-dependent protein kinase belonging to a proposed calcium-triggered signaling cascade involved in a number of cellular processes. Isoform 1, isoform 2 and isoform 3 phosphorylate CAMK1 and CAMK4. Isoform 3 phosphorylates CAMK1D. Isoform 4, isoform 5 and isoform 6 lacking part of the calmodulin- binding domain are inactive. Seems to be involved in hippocampal activation of CREB1. Belongs to the protein kinase superfamily. Ser/Thr protein kinase family. 7 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Mouse CAMKK2 camkk2 (Catalog #AAA7237822) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7237822 ELISA Kit recognizes Mouse CAMKK2. It is sometimes possible for the material contained within the vial of "Calcium/calmodulin-dependent protein kinase kinase 2 (CAMKK2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
