Intended Uses: This CKAP5 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat CKAP5. This ELISA kit for research use only!

Principle of the Assay: CKAP5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CKAP5 antibody and an CKAP5-HRP conjugate. The assay sample and buffer are incubated together with CKAP5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CKAP5 concentration since CKAP5 from samples and CKAP5-HRP conjugate compete for the anti-CKAP5 antibody binding site. Since the number of sites is limited, as more sites are occupied by CKAP5 from the sample, fewer sites are left to bind CKAP5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CKAP5 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a cytoskeleton-associated protein which belongs to the TOG/XMAP215 family. The N-terminal half of this protein contains a microtubule-binding domain and the C-terminal half contains a KXGS motif for binding tubulin dimers. This protein has two distinct roles in spindle formation; it protects kinetochore microtubules from depolymerization and plays an essential role in centrosomal microtubule assembly. This protein may be necessary for the proper interaction of microtubules with the cell cortex for directional cell movement. It also plays a role in translation of the myelin basic protein (MBP) mRNA by interacting with heterogeneous nuclear ribonucleoprotein (hnRNP) A2, which associates with MBP. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Aug 2011]
