Human Cysteine protease ATG4C (ATG4C) ELISA Kit
Principle of the Assay: ATG4C ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ATG4C antibody and an ATG4C-HRP conjugate. The assay sample and buffer are incubated together with ATG4C-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATG4C concentration since ATG4C from samples and ATG4C-HRP conjugate compete for the anti-ATG4C antibody binding site. Since the number of sites is limited, as more sites are occupied by ATG4C from the sample, fewer sites are left to bind ATG4C-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATG4C concentration in each sample is interpolated from this standard curve.

Autophagy is postulated to be essential for cell homeostasis and cell remodeling during differentiation, metamorphosis, non-apoptotic cell death, and aging. Reduced levels of autophagy have been described in some malignant tumors, and a role for autophagy in controlling the unregulated cell growth linked to cancer has been proposed. This gene encodes a member of the autophagin protein family. The encoded protein is also designated as a member of the C-54 family of cysteine proteases. ATG4C: Cysteine protease required for autophagy, which cleaves the C-terminal part of either MAP1LC3, GABARAPL2 or GABARAP, allowing the liberation of form I. A subpopulation of form I is subsequently converted to a smaller form (form II). Form II, with a revealed C-terminal glycine, is considered to be the phosphatidylethanolamine (PE)-conjugated form, and has the capacity for the binding to autophagosomes. Highly expressed in skeletal muscle, heart, liver and testis. Inhibited by N-ethylmaleimide. Belongs to the peptidase C54 family.
