Human beta enolase (ENO3) ELISA Kit
Principle of the assay: ENO3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ENO3 antibody and an ENO3-HRP conjugate. The assay sample and buffer are incubated together with ENO3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ENO3 concentration since ENO3 from samples and ENO3-HRP conjugate compete for the anti-ENO3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ENO3 from the sample, fewer sites are left to bind ENO3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ENO3 concentration in each sample is interpolated from this standard curve.

ENO3: Appears to have a function in striated muscle development and regeneration. Defects in ENO3 are the cause of glycogen storage disease type 13 (GSD13). A metabolic disorder that results in exercise-induced myalgias, generalized muscle weakness and fatigability. It is characterized by increased serum creatine kinase and decreased enolase 3 activity. Dramatically reduced protein levels with focal sarcoplasmic accumulation of glycogen- beta particles are detected on ultrastructural analysis. Belongs to the enolase family. 3 isoforms of the human protein are produced by alternative splicing.
