Principle of the Assay: CNN3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CNN3 antibody and an CNN3-HRP conjugate. The assay sample and buffer are incubated together with CNN3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CNN3 concentration since CNN3 from samples and CNN3-HRP conjugate compete for the anti-CNN3 antibody binding site. Since the number of sites is limited, as more sites are occupied by CNN3 from the sample, fewer sites are left to bind CNN3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CNN3 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a protein with a markedly acidic C terminus; the basic N-terminus is highly homologous to the N-terminus of a related gene, CNN1. Members of the CNN gene family all contain similar tandemly repeated motifs. This encoded protein is associated with the cytoskeleton but is not involved in contraction. Uniprot Description
calponin 3: Thin filament-associated protein that is implicated in the regulation and modulation of smooth muscle contraction. It is capable of binding to actin, calmodulin, troponin C and tropomyosin. The interaction of calponin with actin inhibits the actomyosin Mg-ATPase activity. Belongs to the calponin family.
Protein type: Motility/polarity/chemotaxis; Cytoskeletal
Chromosomal Location of Human Ortholog: 1p22-p21
Cellular Component: focal adhesion; cytoplasm; actin cytoskeleton
Molecular Function: calmodulin binding; actin binding
Biological Process: epithelial cell differentiation; actomyosin structure organization and biogenesis

Product Notes
The Human CNN3 cnn3 (Catalog #AAA7210862) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7210862 ELISA Kit recognizes Human CNN3. It is sometimes possible for the material contained within the vial of "Calponin 3 (CNN3), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
