Intended Uses: This DBI ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine DBI. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: DBI ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-DBI antibody and an DBI-HRP conjugate. The assay sample and buffer are incubated together with DBI-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DBI concentration since DBI from samples and DBI-HRP conjugate compete for the anti-DBI antibody binding site. Since the number of sites is limited, as more sites are occupied by DBI from the sample, fewer sites are left to bind DBI-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DBI concentration in each sample is interpolated from this standard curve.

This gene encodes diazepam binding inhibitor, a protein that is regulated by hormones and is involved in lipid metabolism and the displacement of beta-carbolines and benzodiazepines, which modulate signal transduction at type A gamma-aminobutyric acid receptors located in brain synapses. The protein is conserved from yeast to mammals, with the most highly conserved domain consisting of seven contiguous residues that constitute the hydrophobic binding site for medium- and long-chain acyl-Coenzyme A esters. Diazepam binding inhibitor is also known to mediate the feedback regulation of pancreatic secretion and the postprandial release of cholecystokinin, in addition to its role as a mediator in corticotropin-dependent adrenal steroidogenesis. Three pseudogenes located on chromosomes 6, 8 and 16 have been identified. Multiple transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Jul 2008]

It is also able to displace diazepam from the benzodiazepine (BZD) recognition site located on the GABA type A receptor. Belongs to the ACBP family. 6 isoforms of the human protein are produced by alternative promoter.
