Intended Uses: This CLIC2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CLIC2. This ELISA kit for research use only!

Principle of the Assay: CLIC2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CLIC2 antibody and an CLIC2-HRP conjugate. The assay sample and buffer are incubated together with CLIC2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLIC2 concentration since CLIC2 from samples and CLIC2-HRP conjugate compete for the anti-CLIC2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CLIC2 from the sample, fewer sites are left to bind CLIC2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLIC2 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a chloride intracellular channel protein. Chloride channels are a diverse group of proteins that regulate fundamental cellular processes including stabilization of cell membrane potential, transepithelial transport, maintenance of intracellular pH, and regulation of cell volume. This protein may play a role in inhibiting the function of ryanodine receptor 2. A mutation in this gene is the cause of X-linked mental retardation-32. [provided by RefSeq, Aug 2013]

Uniprot Description

CLIC2: Can insert into membranes and form chloride ion channels. Channel activity depends on the pH. Membrane insertion seems to be redox-regulated and may occur only under oxydizing conditions. Modulates the activity of RYR2 and inhibits calcium influx. Belongs to the chloride channel CLIC family.

Product Notes

The Human CLIC2 clic2 (Catalog #AAA7203397) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7203397 ELISA Kit recognizes Human CLIC2. It is sometimes possible for the material contained within the vial of "Chloride intracellular channel protein 2 (CLIC2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
