Determined by its ability to chemoattract human blood monocytes using a concentration range of 5.0-20.0 ng/ml.

Both MIP-1alpha and MIP-1beta are structurally and functionally related CC chemokines. They participate in the host response to invading bacterial, viral, parasite and fungal pathogens by regulating the trafficking and activation state of selected subgroups of inflammatory cells e.g. macrophages, lymphocytes and NK cells. While both MIP-1alpha and MIP-1beta exert similar effects on monocytes their effect on lymphocytes differ; with MIP-1alpha selectively attracting CD8+ lymphocytes and MIP-1beta selectively attracting CD4+ lymphocytes. Additionally, MIP-1alpha and MIP-1beta have also been shown to be potent chemoattractants for B cells, eosinophils and dendritic cells. Both human and murine MIP-1alpha and MIP-1beta are active on human and murine hematopoietic cells. Recombinant human MIP-1betais a 7.6 kDa protein containing 69 amino acid residues, including the four highly conserved cysteine residues present in CC chemokines.

NCBI Description

This gene is one of several cytokine genes clustered on the q-arm of chromosome 17. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. This protein is similar to CCL4 which inhibits HIV entry by binding to the cellular receptor CCR5. The copy number of this gene varies among individuals; most individuals have 1-5 copies in the diploid genome, although rare individuals do not contain this gene at all. The human genome reference assembly contains two copies of this gene. This record represents the more centromeric gene. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Monokine with inflammatory and chemokinetic properties. Binds to CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant MIP-1-beta induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form MIP-1-beta(3-69) retains the abilities to induce down-modulation of surface expression of the chemokine receptor CCR5 and to inhibit the CCR5-mediated entry of HIV-1 in T-cells. MIP-1-beta(3-69) is also a ligand for CCR1 and CCR2 isoform B Ref.13 Ref.14 Ref.15

Subunit structure: Homodimer and heterodimer of MIP-1-alpha(4-69) and MIP-1-beta(3-69). Ref.15

Subcellular location: Secreted.

Induction: By mitogens.

Post-translational modification: N-terminal processed form MIP-1-beta(3-69) is produced by proteolytic cleavage after secretion from peripheral blood lymphocytes.

Sequence similarities: Belongs to the intercrine beta (chemokine CC) family.

Caution: Was originally (Ref.12) thought to be a ligand for CCR8.
