Supplied as a lyophilized powder from 10mM Tris pH 8.0 and 25mM sodium chloride. Reconstitute with sterile 5mM sodium citrate, pH 2.8, to a concentration of 0.1-1mg/ml. Slow to dissolve. Do not vortex.
Biological Activity
The ED50 was determined by the dose-dependent proliferation of the RPMI 8226 cells. The expected ED50 for this effect is 15.0-20.0ng/ml.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Reconstitute with sterile 5mM sodium citrate, pH 2.8. For extended storage, add carrier protein (e.g., 0.1% BSA). Aliquot and store at -20 degree C. Reconstituted product is stable for 3 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Visfatin is a 55kD protein produced and secreted primarily by white adipose tissue. Recently, Visfatin was isolated from visceral fat deposits and shown to possess insulin-mimetic activity. Like insulin, Visfatin exerts hypoglycemic effects by interacting with the insulin receptor. The binding affinity of Visfatin for the insulin receptor is similar to that of insulin, but it does not compete with insulin, suggesting that the two proteins interact with different receptor sites.

NCBI Description

This gene encodes a protein that catalyzes the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, one step in the biosynthesis of nicotinamide adenine dinucleotide. The protein belongs to the nicotinic acid phosphoribosyltransferase (NAPRTase) family and is thought to be involved in many important biological processes, including metabolism, stress response and aging. This gene has a pseudogene on chromosome 10. [provided by RefSeq]

Uniprot Description

Function: Catalyzes the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, an intermediate in the biosynthesis of NAD. It is the rate limiting component in the mammalian NAD biosynthesis pathway

Catalytic activity: Nicotinamide D-ribonucleotide + diphosphate = nicotinamide + 5-phospho-alpha-D-ribose 1-diphosphate.

Enzyme regulation: Inhibited by FK866. FK866 competes for the same binding site as nicotinamide, but due to its very low dissociation rate, it is essentially an irreversible inhibitor. Ref.11

Pathway: Cofactor biosynthesis; NAD(+) biosynthesis; nicotinamide D-ribonucleotide from 5-phospho-alpha-D-ribose 1-diphosphate and nicotinamide: step 1/1.

Subunit structure: Homodimer. Ref.11

Subcellular location: Cytoplasm

Tissue specificity: Expressed in large amounts in bone marrow, liver tissue, and muscle. Also present in heart, placenta, lung, and kidney tissues.

Sequence similarities: Belongs to the NAPRTase family.

Caution: Was originally (Ref.1) thought to be a cytokine which acts on early B-lineage precursor cells, by enhancing the effect of IL-7 and SCF on pre-B-cell colony formation.
