FLISA: 1:1,000
WB: 1:100-1:500
Applications are based on unconjugated antibody.
Western Blot (WB)
(The anti-CDKN1A Pab is used in Western blot to detect CDKN1A in T-47D cell lysate.)
Immunohistochemistry (IHC)
(Formalin-fixed and paraffin-embedded human cancer tissue reacted with the primary antibody, which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated. BC = breast carcinoma; HC = hepatocarcinoma.)
Immunofluorescence (IF)
(Confocal immunofluorescent analysis of p21 (CDKN1A) Antibody (C-term) with 293 cell followed by Alexa Fluor 488-conjugated goat anti-rabbit lgG (green).Actin filaments have been labeled with Alexa Fluor 555 phalloidin (red).DAPI was used to stain the cell nuclear (blue).)
Flow Cytometry (FC/FACS)
(p21 (CDKN1A) Antibody (C-term) flow cytometric analysis of 293 cells (right histogram) compared to a negative control cell (left histogram).FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.)
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a potent cyclin-dependent kinase inhibitor. The encoded protein binds to and inhibits the activity of cyclin-CDK2 or -CDK4 complexes, and thus functions as a regulator of cell cycle progression at G1. The expression of this gene is tightly controlled by the tumor suppressor protein p53, through which this protein mediates the p53-dependent cell cycle G1 phase arrest in response to a variety of stress stimuli. This protein can interact with proliferating cell nuclear antigen (PCNA), a DNA polymerase accessory factor, and plays a regulatory role in S phase DNA replication and DNA damage repair. This protein was reported to be specifically cleaved by CASP3-like caspases, which thus leads to a dramatic activation of CDK2, and may be instrumental in the execution of apoptosis following caspase activation. Multiple alternatively spliced variants have been found for this gene. [provided by RefSeq, Nov 2010]
Uniprot Description

Product Notes
The p21 cdkn1a (Catalog #AAA6491316) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The p21, CT (Cyclin-dependent Kinase Inhibitor 1, CDK-interacting Protein 1, Melanoma Differentiation-associated Protein 6, MDA-6, CDKN1A, CAP20, CDKN1, CIP1, MDA6, PIC1, SDI1, WAF1) (PE) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's p21 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC), FLISA, Western Blot (WB). IHC: 1:50-1:100 FLISA: 1:1,000 WB: 1:100-1:500 Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the p21 cdkn1a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "p21, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
