Related Product Information for anti-Sirt5 antibody
This gene encodes a member of the sirtuin family of proteins, homologs to the yeast Sir2 protein. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class III of the sirtuin family. Alternative splicing of this gene results in multiple transcript variants.

Uniprot Description

Function: NAD-dependent lysine demalonylase and desuccinylase that specifically removes malonyl and succinyl groups on target proteins. Activates CPS1 and contributes to the regulation of blood ammonia levels during prolonged fasting: acts by mediating desuccinylation of CPS1, thereby increasing CPS1 activity in response to elevated NAD levels during fasting. Has weak NAD-dependent protein deacetylase activity; however this activity may not be physiologically relevant in vivo. Can deacetylate cytochrome c (CYCS) and a number of other proteins in vitro. Ref.2 Ref.3 Ref.4

Catalytic activity: NAD+ + a malonylprotein = nicotinamide + O-malonyl-ADP-ribose + a protein. HAMAP-Rule MF_03160NAD+ + a succinylprotein = nicotinamide + O-succinyl-ADP-ribose + a protein. HAMAP-Rule MF_03160

Cofactor: Binds 1 zinc ion per subunit

By similarity. HAMAP-Rule MF_03160

Subunit structure: Monomer. Homodimer. Interacts with CPS1. Ref.2

Subcellular location: Mitochondrion Ref.2.

Tissue specificity: Detected in brain, liver, heart, kidney, lung, thymus, spleen, skeletal muscle, intestine, pancreas and testis (at protein level). Ref.2

Domain: In contrast to class I sirtuins, class III sirtuins have only weak deacetylase activity. Difference in substrate specificity is probably due to a larger hydrophobic pocket with 2 residues (Tyr-102 and Arg-105) that bind to malonylated and succinylated substrates and define the specificity

By similarity. HAMAP-Rule MF_03160

Disruption phenotype: No visible phenotype. After 48 hours of fasting, the absence of Cps1 activation leads to elevated blood ammonia levels, possibly due to the presence of succinylation at 'Lys-1291' in Cps1. Ref.2 Ref.3 Ref.4

Sequence similarities: Belongs to the sirtuin family. Class III subfamily.Contains 1 deacetylase sirtuin-type domain.

Product Notes

The Sirt5 sirt5 (Catalog #AAA647773) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The SIRT5, ID (SIRT5, SIR2L5, NAD-dependent protein deacylase sirtuin-5, mitochondrial, Regulatory protein SIR2 homolog 5, SIR2-like protein 5) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's SIRT5 can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB). Suitable for use in Western Blot, ELISA Dilution: ELISA: 1:1,000 Western Blot: 1:100-500. Researchers should empirically determine the suitability of the Sirt5 sirt5 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SIRT5, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
