Rabbit anti-Human FGFR4 Polyclonal Antibody | anti-FGFR4 antibody
FGFR4, NT (FGFR4, JTK2, TKF, Fibroblast growth factor receptor 4, CD334) (PE)
Western Blot (WB)
(Western Blot analysis in 293 cell line lysates (35ug/lane) usingMBS6012430. This demonstrates that MBS6012430 detected the FGFR4 protein (arrow).)
Immunohistochemistry (IHC)
(Immunohistochemistry analysis in formalin-fixed and paraffin-embedded human cancer tissue reacted with theMBS6012430, which was peroxidase-conjugated to the secondary antibody, followed by AEC staining.)
Flow Cytometry (FC/FACS)
(Flow Cytometric analysis of WiDr cells using MBS6012430 (bottom histogram) compared to a negative control cell (top histogram). FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.)
Immunofluorescence (IF)
(Fluorescent confocal image of HeLa cells stained withMBS6012430. HeLa cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with MBS6012430 (1:200, 2 hours at RT). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1 hour). Nuclei were counterstained with Hoechst 33342 (blue) (10ug/ml, 5 min). Note the highly specific localization of the FGFR4 mainly to the nucleus, supported by Human Protein Atlas Data)
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a member of the fibroblast growth factor receptor family, where amino acid sequence is highly conserved between members and throughout evolution. FGFR family members differ from one another in their ligand affinities and tissue distribution. A full-length representative protein would consist of an extracellular region, composed of three immunoglobulin-like domains, a single hydrophobic membrane-spanning segment and a cytoplasmic tyrosine kinase domain. The extracellular portion of the protein interacts with fibroblast growth factors, setting in motion a cascade of downstream signals, ultimately influencing mitogenesis and differentiation. The genomic organization of this gene, compared to members 1-3, encompasses 18 exons rather than 19 or 20. Although alternative splicing has been observed, there is no evidence that the C-terminal half of the IgIII domain of this protein varies between three alternate forms, as indicated for members 1-3. This particular family member preferentially binds acidic fibroblast growth factor and, although its specific function is unknown, it is overexpressed in gynecological tumor samples, suggesting a role in breast and ovarian tumorigenesis. Uniprot Description
FGFR4: a tyrosine-kinase receptor that acts as cell-surface receptor for fibroblast growth factors and plays a role in the regulation of cell proliferation, differentiation and migration, and in regulation of lipid metabolism, bile acid biosynthesis, glucose uptake, vitamin D metabolism and phosphate homeostasis. Required for normal down-regulation of the expression of CYP7A1, the rate-limiting enzyme in bile acid synthesis, in response to FGF19. Phosphorylates PLCG1 and FRS2. Ligand binding leads to the activation of several signaling cascades. Activation of PLCG1 leads to the production of the cellular signaling molecules diacylglycerol and inositol 1,4,5-trisphosphate. Phosphorylation of FRS2 triggers recruitment of GRB2, GAB1, PIK3R1 and SOS1, and mediates activation of the MAP kinase and AKT signaling pathways. Promotes SRC-dependent phosphorylation of the matrix protease MMP14 and its lysosomal degradation. FGFR4 signaling is down-regulated by receptor internalization and degradation; MMP14 promotes internalization and degradation of FGFR4. Mutations that lead to constitutive kinase activation or impair normal FGFR4 inactivation lead to aberrant signaling. Two isoforms of the human protein are formed by alternative splicing. Product Notes
The FGFR4 fgfr4 (Catalog #AAA6299186) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The FGFR4, NT (FGFR4, JTK2, TKF, Fibroblast growth factor receptor 4, CD334) (PE) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's FGFR4 can be used in a range of immunoassay formats including, but not limited to, FLISA, Western Blot (WB), Immunohistochemistry (IHC), Flow Cytometry (FC/FACS), Immunofluorescence (IF). Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the FGFR4 fgfr4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "FGFR4, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
