The Hey family of bHLH transcription factors are direct targets of Notch signaling. HEY2 (gridlock) is an important determinant of mammalian heart development, loss of which in mouse leads to cardiac defects with high postnatal lethality. Mouse Hey1 knockouts have no apparent phenotypic defect but the combined loss of Hey1 and Hey2 results in embryonic death after embryonic day 9.5 (E9.5) with a global lack of vascular remodeling and massive hemorrhage. Similar defects are observed in Jagged1 and Notch1 knockout mice.

This gene encodes a protein that contains a SET domain. SET domains appear to be protein-protein interaction domains that mediate interactions with a family of proteins that display similarity with dual-specificity phosphatases (dsPTPases). The function of this gene has not been determined. Two alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Jul 2008]

SUV420H1: a protein methyltransferase that specifically trimethylates 'Lys-20' of histone H4. H4 'Lys-20' trimethylation represents a specific tag for epigenetic transcriptional repression. Mainly functions in pericentric heterochromatin regions, thereby playing a central role in the establishment of constitutive heterochromatin in these regions. SUV420H1 is targeted to histone H3 via its interaction with RB1 family proteins (RB1, RBL1 and RBL2). Strongly down-regulated in breast cancer cells. Belongs to the histone-lysine methyltransferase family. Suvar4-20 subfamily. Three alternatively-spliced isoforms have been described.
