Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Yotiao is a novel cytoskeletal protein specifically concentrated in the neuromuscular junction and neuronal synapes that interacts with specific splice variants of NMDA Receptor Subunit NR1 in a C1 exon cassette-dependent manner (1). Yotiao has also been shown to physically attach the type I protein phosphatase (PP1) and the adenosine 3',5'-mono-phosphate (cAMP)- dependent protein kinase (PKA) holoenzyme to NMDA receptors to regulate channel activity (2). NMDA receptors are involved with many important functions and dysfunctions of the nervous system, including synapse formation, synaptic plasticity, and excitotoxicity. Yotiao research may lead to a clearer understanding of these neural processes by explaining how NMDA receptors are targeted to particular synapses, how this localization is regulated during development and synaptic activity, and how NMDA receptor activity is transduced into intracellular signals responsible for particular neuronal responses (1). Human tissue distribution studies with yotiao mRNA revealed that yotiao is abundantly present in skeletal muscle and pancreas, with lower expression in heart, placenta, and brain. Yotiao mRNA was undetectable in lung, liver, and kidney (1).

The A-kinase anchor proteins (AKAPs) are a group of structurally diverse proteins which have the common function of binding to the regulatory subunit of protein kinase A (PKA) and confining the holoenzyme to discrete locations within the cell. This gene encodes a member of the AKAP family. Alternate splicing of this gene results in at least two isoforms that localize to the centrosome and the Golgi apparatus, and interact with numerous signaling proteins from multiple signal transduction pathways. These signaling proteins include type II protein kinase A, serine/threonine kinase protein kinase N, protein phosphatase 1, protein phosphatase 2a, protein kinase C-epsilon and phosphodiesterase 4D3. Uniprot Description

AKAP9: Binds to type II regulatory subunits of protein kinase A. Scaffolding protein that assembles several protein kinases and phosphatases on the centrosome and Golgi apparatus. May be required to maintain the integrity of the Golgi apparatus. Isoform 4 is associated with the N-methyl-D-aspartate receptor and is specifically found in the neuromuscular junction (NMJ) as well as in neuronal synapses, suggesting a role in the organization of postsynaptic specializations. Defects in AKAP9 are the cause of long QT syndrome type 11 (LQT11). Long QT syndromes are heart disorders characterized by a prolonged QT interval on the ECG and polymorphic ventricular arrhythmias. They cause syncope and sudden death in response to excercise or emotional stress. They can present with a sentinel event of sudden cardiac death in infancy. 6 isoforms of the human protein are produced by alternative splicing.
