Adipocytes synthesize and store energy in periods of nutritional abundance and mobilize lipids during starvation and other times of need. In order to accomplish these complex tasks energy balance, adipocytes express many genes, including Pref-1, involved in lipid metabolism and glucose homeostasis. Preadipocyte factor-1 (Pref-1), an epidermal growth factor-like domain-containing transmembrane protein is implicated in inhibiting preadipocytes differentiation. Pref-1 (mouse 385aa, rat 383aa, human 383aa, chromosome 14q32), also known as fetal antigen 1 (FA1), Delta Drosophila homolog-like 1 (DLK1) or Zona glomerulosa-specific factor (ZOG), is synthesized as a membrane protein in preadipocytes but it is not detectable in mature adipocytes. Dexamethasone, a promoter of adipocyte differentiation, down regulates Pref-1. Pref-1 is proteolytically cleaved to generate a biologically active 50kD ectodomain (24-303aa) or soluble form. Constitutive expression of Pref-1 or addition of its ectodomain inhibits adipogenesis. There are at least four major isoforms (45-60kD) of Pref-1 generated by alternatively splicing. Pref-1/DLK is expressed in tumors with neuroendocrine features, such as neuroblastoma, pheochromocytoma, and a subset of small cell lung carcinoma cell lines. Its expression in normal tissues is restricted to the adrenal gland and placenta.

NCBI Description

This gene encodes a transmembrane protein containing six epidermal growth factor repeats. The protein is involved in the differentiation of several cell types, including adipocytes; it is also thought to be a tumor suppressor. It is one of several imprinted genes located in a region of on chr 14q32. Certain mutations in this imprinted region can cause phenotypes similar to maternal and paternal uniparental disomy of chromosome 14 (UPD14). This gene is expressed from the paternal allele. A polymorphism within this gene has been associated with child and adolescent obesity. The mode of inheritance for this polymorphism is polar overdominance; this non-Mendelian inheritance pattern was first described in sheep with the callipyge phenotype, which is characterized by muscle hypertrophy and decreased fat mass.
