Application Notes
Suitable for use in Western Blot, Immunohistochemistry and ELISA., , Recommended Dilutions:, Western Blot: 0.1-0.2ug/ml The detection limit is ~1ng/lane and 5ng/lane under non-reducing and reducing conditions, respectively., Immunohistochemistry: 10ug/ml detects DMP1 in E15.5 mouse embryos. Fix cells with PBS, 4% paraformaldehyde for 20 minutes at RT and block with PBS, 10% normal donkey serum, 0.1% Triton X-100, 1% BSA for 45 minutes at RT. Incubate cells with D8075-51 for 3 hours at RT followed by donkey anti-sheep lgG at RT for 1 hour. Between each step, wash cells with PBS, 0.1% BSA., Direct ELISA: 0.5-1.0ug/ml The detection limit is ~0.1ng/well., Optimal dilutions to be determined by the researcher.

NSO myeloma cells with spleen cells from Balb/c mice.

Storage and Stability:
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

DMP-1 (dentin matrix protein 1) is an acidic, 100-160kD member of the phosphophoryn family of extracellular matrix proteins. Mouse DMP-1 is found both intra- and extracellularly, where phosphorylated DMP-1 participates in bone mineralization. Secreted mature mouse DMP-1 is a 487 amino acid (aa) proteoglycan. It contains multiple serine, glutamic acid, and aspartic acid residues, a GAG attachment site (aa71-72) and an RGD motif (aa350-352). Multiple cleavage sites exist between aa205-253, creating multiple fragments ~37kD and 57kD in size.

Uniprot Description

DMP1: May have a dual function during osteoblast differentiation. In the nucleus of undifferentiated osteoblasts, unphosphorylated form acts as a transcriptional component for activation of osteoblast-specific genes like osteocalcin. During the osteoblast to osteocyte transition phase it is phosphorylated and exported into the extracellular matrix, where it regulates nucleation of hydroxyapatite. Defects in DMP1 are the cause of rickets hypophosphatemic autosomal recessive type 1 (ARHR1). A hereditary form of hypophosphatemic rickets, a disorder of proximal renal tubule function that causes phosphate loss, hypophosphatemia and skeletal deformities, including rickets and osteomalacia unresponsive to vitamin D. Symptoms are bone pain, fractures and growth abnormalities. 2 isoforms of the human protein are produced by alternative splicing.
