Dilution: Western Blot: 1:2000 detects phospho-estrogen receptor (Ser118) in RIPA lysates from MCF-7 cells treated with estradiol and EGF, and recombinant estrogen receptor phosphorylated in vitro with MAP Kinase 2/Erk2, active. Lysates from estradiol and EGF treated MCF-7 cells (lanes 1 and 2) or recombinant estrogen receptor protein phosphorylated with MAPK2/Erk2 (lanes 3 and 4) were resolved by electrophoresis and transferred to nitrocellulose. Following blocking of the membrane, the nitrocellulose was either treated with lambda phosphatase (lanes 1 and 3) or left untreated (lanes 2 and 4), then probed with anti-phospho- Estrogen Receptor (Ser118) (1:2000 dilution). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher.

This gene encodes an estrogen receptor, a ligand-activated transcription factor composed of several domains important for hormone binding, DNA binding, and activation of transcription. The protein localizes to the nucleus where it may form a homodimer or a heterodimer with estrogen receptor 2. Estrogen and its receptors are essential for sexual development and reproductive function, but also play a role in other tissues such as bone. Estrogen receptors are also involved in pathological processes including breast cancer, endometrial cancer, and osteoporosis.
