Application Notes
Suitable for use in ELISA and Western Blot.
Dilution: Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~20ng/well.
Western Blot: 1-2ug/ml detects human CNP peptides containing aa98-126. Using a colormetric system, the detection limit is ~25ng/lane under non-reducing and reducing conditions.
Immunogen
E. coli-derived recombinant protein corresponding to aa24-126 from human CNP precursor (Accession #P23582). Species sequence homology: mouse and rat.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

C-Type Natriuretic Peptide (CNP) belongs to the natriuretic peptide family and functions in an autocrine or paracrine fashion. CNP interacts with the GC-B/NPR-B receptor to promote vasorelaxation, vascular remodeling, and the growth and differentiation of bone and neural tissue. CNP is synthesized as a prohormone that is cleaved intracellularly by furin, yielding a 49aa propeptide and a 53aa mature peptide. Additional cleavage of the mature peptide generates 29aa and 22aa peptides which have comparable activity in some assays. Within the 53aa mature CNP, human shares 96% aa sequence identity with mouse and rat.
