Related Product Information for anti-SPP1 antibody
Osteopontin is a single-chain polypeptide with a molecular weight of ~35kD. It is a glycoprotein first identified in osteoblasts. It is biosynthesized by a variety of tissue types including preosteoblasts, osteoblasts, osteocytes, extraosseous cells in the inner ear, brain, kidney, deciduum, placenta, odontoblasts, some bone marrow cells, hypertrophic chondrocytes, macrophages, smooth muscle, and endothelial cells. Osteopontin is overexpressed in a variety of cancers, including lung cancer, breast cancer, colorectal cancer, stomach cancer, ovarian cancer, melanoma and mesothelioma. It may contribute to kidney stone formation and both glomerulonephritis and tubulointerstitial nephritis and is also found in atheromatous plaques within arteries. Research has implicated osteopontin in excessive scar-forming and a gel has been developed to inhibit its effect.

NCBI Description

The protein encoded by this gene is involved in the attachment of osteoclasts to the mineralized bone matrix. The encoded protein is secreted and binds hydroxyapatite with high affinity. The osteoclast vitronectin receptor is found in the cell membrane and may be involved in the binding to this protein. This protein is also a cytokine that upregulates expression of interferon-gamma and interleukin-12. Several transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2011]

Uniprot Description

osteopontin: Binds tightly to hydroxyapatite. Appears to form an integral part of the mineralized matrix. Probably important to cell-matrix interaction. Belongs to the osteopontin family. 5 isoforms of the human protein are produced by alternative splicing.

Protein type: Secreted; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 4q22.1

Cellular Component: extracellular space; cell projection; apical part of cell; perinuclear region of cytoplasm; extracellular region

Molecular Function: protein binding; extracellular matrix binding; cytokine activity

Biological Process: osteoblast differentiation; neutrophil chemotaxis; extracellular matrix disassembly; extracellular matrix organization and biogenesis; response to vitamin D; biomineral formation; response to steroid hormone stimulus; negative regulation of collateral sprouting of intact axon in response to injury; positive regulation of bone resorption; decidualization; inflammatory response; cell adhesion; embryo implantation

Product Notes

The SPP1 spp1 (Catalog #AAA6000966) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Osteopontin, aa75-90 (OPN, Bone Sialoprotein I, BSP-1, Secreted Phosphoprotein 1, SPP1, Early T-Lymphocyte Activation 1) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Osteopontin, aa75-90 can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Immunohistochemistry (IHC). Suitable for use in ELISA, Immunohistochemistry and Western Blot. Dilution: ELISA: 1:1000 Immunohistochemistry (Paraffin): 1:1500 Western Blot: 1:500 detected Osteopontin in 10ug of RAW 264.7 lysates. Researchers should empirically determine the suitability of the SPP1 spp1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Osteopontin, aa75-90, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
