Precipitation assays. In immunoelectrophoresis use 2 ul or equivalent against 120 ul antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 ul in a 3 mm diameter center well and 2 ul serum samples (neat and serially diluted) in 2 mm diameter peripheral wells.

Alpha-2 ceruloplasmin is copper containing alpha-2 glycoprotein with a molecular weight of 160,000. Its mean concentration in plasma of adults is 32 mg/100ml. Elevated levels are observed in pregnant women, in patients with infectious diseases and the nephritic syndrome. More than 90% of the plasma copper is bound to this protein. Ceruloplasmin is isolated from pooled human serum by sequential precipitation and purified by ion exchange chromatography and affinity chromatography. Freund’s complete adjuvant is used in the first step of the immunization procedure.

Delipidated, heat inactivated, lyophilized, stable whole antiserum No preservative added. Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added. Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water.
Preparation and Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.

The reactivity of the antiserum is restricted to alpha-2 ceruloplasmin. In immunoelectrophoresis and radial immunodiffusion (Ouchterlony), using various antiserum concentrations against fresh normal human plasma a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified alpha-2 ceruloplasmin. No reaction is obtained with any other plasma protein component or serum. In precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion to identify the presence of alpha-2 ceruloplasmin in human plasma or other body fluids or to determine its concentration. The presence of non precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in more sensitive non-precipitating antibody-binding techniques if proper controls are included.

NCBI Description

The protein encoded by this gene is a metalloprotein that binds most of the copper in plasma and is involved in the peroxidation of Fe(II)transferrin to Fe(III) transferrin. Mutations in this gene cause aceruloplasminemia, which results in iron accumulation and tissue damage, and is associated with diabetes and neurologic abnormalities. Two transcript variants, one protein-coding and the other not protein-coding, have been found for this gene. [provided by RefSeq, Feb 2012]

CP: Ceruloplasmin is a blue, copper-binding (6-7 atoms per molecule) glycoprotein. It has ferroxidase activity oxidizing Fe(2+) to Fe(3+) without releasing radical oxygen species. It is involved in iron transport across the cell membrane. Provides Cu(2+) ions for the ascorbate-mediated deaminase degradation of the heparan sulfate chains of GPC1. May also play a role in fetal lung development or pulmonary antioxidant defense. Defects in CP are the cause of aceruloplasminemia (ACERULOP). It is an autosomal recessive disorder of iron metabolism characterized by iron accumulation in the brain as well as visceral organs. Clinical features consist of the triad of retinal degeneration, diabetes mellitus and neurological disturbances. Ceruloplasmin levels are decreased in Wilson disease, in which copper cannot be incorporated into ceruloplasmin in liver because of defects in the copper-transporting ATPase 2. Belongs to the multicopper oxidase family.

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ceruloplasmin, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage.
