Application Notes
Applications: Immunoprecipitation. In immunoelectrophoresis use 2 ul human serum against 120 ul antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 ul antiserum in a 3 mm diameter center well and 10 ul of the antiserum in 3 mm diameter peripheral wells. In single radial immunodiffusion use 1% antiserum in the gel.

Properdin is a factor of the alternative complement activation pathway. It has a molecular weight of 222,000 and consists of four peptide chains. Properdin is stabilizing the C3 convertase C3bBb. The mean concentration in plasma is 20 mg/ml. The antigen has been isolated form pooled human plasma. Freund’s complete adjuvant is used in the first step of the immunization procedure.

Delipidated, heat inactivated, lyophilized, stable whole antiserum No preservative added. Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added.
Reactivity Note
The antiSerum does not cross react with any other component of Human plasma. Inter- species cross-reactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. Cross-reactivity of this antiSerum has not been tested in detail.
Preparation and Storage
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4 degree C; prolonged storage at or below -20 degree C. Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4 degree C, not refrozen, and preferably used the same day. Lyophilized at +4 degree C--at least 10 years. Reconstituted at or below -20 degree C--3-5 years. Reconstituted at +4 degree C--7 days

The defined antibody specificity is restricted to properdin as tested at the level of sensitivity of immunoprecipitation techniques. A single precipitin line is obtained in immuno- electrophoresis, bidimensional electrophoresis and double radial immunodiffusion (Ouchterlony) against normal plasma, which shows a reaction of identity with the precipitated purified properdin. No reaction is obtained with any other plasma protein. In precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in non-precipitating antibody-binding techniques if proper controls are included.

NCBI Description

This gene encodes a plasma glycoprotein that positively regulates the alternative complement pathway of the innate immune system. This protein binds to many microbial surfaces and apoptotic cells and stabilizes the C3- and C5-convertase enzyme complexes in a feedback loop that ultimately leads to formation of the membrane attack complex and lysis of the target cell. Mutations in this gene result in two forms of properdin deficiency, which results in high susceptibility to meningococcal infections. Multiple alternatively spliced variants, encoding the same protein, have been identified.[provided by RefSeq, Feb 2009]

CFP: A positive regulator of the alternate pathway of complement. It binds to and stabilizes the C3- and C5-convertase enzyme complexes. Defects in CFP are the cause of properdin deficiency (PFD). PFD results in higher susceptibility to bacterial infections; especially to meningococcal infections. Three phenotypes have been reported: complete deficiency (type I), incomplete deficiency (type II), and dysfunction of properdin (type III).

It is sometimes possible for the material contained within the vial of "properdin, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
