Form/Format
Each vial contains 100 ul 1 mg/ml purified monoclonal antibody in PBS containing 0.09% sodium azide.
Applicable Applications for anti-PKP3 antibody
Immunocytochemistry (IHC), Immunohistochemistry (IHC) (frozen), Immunoprecipitation (IP), Western Blot (WB)
Application Notes
12B11F8 is suitable for immunocytochemistry (methanol-fixed cells), immunohistochemistry on frozen tissues, immunoprecipitation and Western blot detection. For frozen tissues use a PBS buffer containing 0.1 mM CaCl2 and 0.1 mM MgCl2. For optimal results, fixed cells or tissues should be treated with 0.2% Triton-X 100 for 15 min. prior to incubation with primary antibody. Optimal antibody dilution should be determined by titRation; recommended range is 1:25 - 1:100 for immunohistochemistry with avidin-biotinylated Horseradish peroxidase complex (ABC) as detection reagent, and 1:100 - 1:1000 for immunoblotting appliCations.
Source Note
12B11F8 is a Mouse monoclonal IgG1 antibody derived by fusion of Mouse myeloma cells with spleen cells from a Mouse immunized with a synthetic peptide corresponding to amino acid residues 779-793 of Human plakophilin-3 coupled to keyhole limpet hemocyanin. (KLHRDFRAKGYRKED).
Preparation and Storage
Store at 4 degree C, or in small aliquots at -20 degree C.
Related Product Information for anti-PKP3 antibody
Plakophilin-3 is an Armadillo-like protein present in nuclei and desmosomes of epithelial cells. The expression pattern of this protein seems to be largely restricted to epithelial cell types. Plakophilin-3 can be detected along cell borders in a punctuate staining pattern typical for desmosomal proteins. In addition to the desmosomal immunolocalisation, immunostaining was observed as bright nuclear speckles. Thus, like plakophilin-1 and-2, plakophilin-3 displays a dual intracellular localisation in the desmosomal plaque and in the cell nucleus, and therefore is probably involved in signal transduction pathways between the plasma membrane and the nucleus. The Human protein has a predicted molecular mass of 87 kD.

NCBI Description

This gene encodes a member of the arm-repeat (armadillo) and plakophilin gene families. Plakophilin proteins contain numerous armadillo repeats, localize to cell desmosomes and nuclei, and participate in linking cadherins to intermediate filaments in the cytoskeleton. This protein may act in cellular desmosome-dependent adhesion and signaling pathways. [provided by RefSeq]

Uniprot Description

plakophilin 3: a novel armadillo-like protein present in nuclei and desmosomes of epithelial cells and dendritic reticular cells of lymphatic follicles.

Protein type: Cell adhesion

Chromosomal Location of Human Ortholog: 11p15

Cellular Component: desmosome; plasma membrane; intercellular junction; nucleus

Product Notes

The PKP3 pkp3 (Catalog #AAA570133) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Mouse anti Plakophilin-3 reacts with Canine, Human, Mouse, Swine and may cross-react with other species as described in the data sheet. AAA Biotech's Plakophilin-3 can be used in a range of immunoassay formats including, but not limited to, Immunocytochemistry (IHC), Immunohistochemistry (IHC) (frozen), Immunoprecipitation (IP), Western Blot (WB). Researchers should empirically determine the suitability of the PKP3 pkp3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Plakophilin-3, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
