Host Note
Optimized DNA sequence encoding Human HCC-2 (CCL15) mature chain was expressed in Escherichia Coli.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note
Human HCC-2, generated by the proteolytic removal of the signal peptide
and propeptide and has a calculated molecular mass of approximately 10 kDa.

Recombinant HCC-2 is a disulfide-linked monomeric protein consisting of 92 amino acid residue subunits and migrates as an approximately 10 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.
Related Product Information for HCC-2 recombinant protein
CCL15 is a human chemokine which acts predominantly on chemokine receptor CCR1 and to a lesser degree on CCR3. It is a potent chemoattractant for monocytes and lymphocytes in vitro and, after i.p. application, CCL15 induces recruitment neutrophils, monocytes, and lymphocytes into mouse peritoneum. CCL15 is constitutively expressed in the gut and in the liver and was identified to circulate in human plasma. The N terminus of CCL15 was found to be important to exert its full biological activity. HCC-2 shares significant sequence homology with CKb8 and the murine chemokines C10, CCF18/MRP-2, and macrophage inflammatory protein 1g, which all contain six instead of four conserved cysteines.

NCBI Description

This gene is located in a cluster of similar genes in the same region of chromosome 17. These genes encode CC cytokines, which are secreted proteins characterized by two adjacent cysteines. The product of this gene is chemotactic for T cells and monocytes, and acts through C-C chemokine receptor type 1 (CCR1). The proprotein is further processed into numerous smaller functional peptides. Naturally-occurring readthrough transcripts occur from this gene into the downstream gene, CCL14 (chemokine (C-C motif) ligand 14). [provided by RefSeq, Jan 2013]

Uniprot Description

CCL15: Chemotactic factor that attracts T-cells and monocytes, but not neutrophils, eosinophils, or B-cells. Acts mainly via CC chemokine receptor CCR1. Also binds to CCR3. CCL15(22-92), CCL15(25-92) and CCL15(29-92) are more potent chemoattractants than the small-inducible cytokine A15. Belongs to the intercrine beta (chemokine CC) family.

Product Notes

The HCC-2 ccl15 (Catalog #AAA553121) is a Recombinant Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: MKVSVAALSC LMLVAVLGSQ AQFINDAETE LMMSKLPLEN PVVLNSFHFA ADCCTSYISQ SIPCSLMKSY FETSSECSKP GVIFLTKKGR QVCAKPSGPG VQDCMKKLKP YSI. It is sometimes possible for the material contained within the vial of "HCC-2 (CCL15), Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
