Endotoxin

Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note
Native human Interleukin-15 is generated by the proteolytic removal of the signal peptide and propeptide, the molecule has a calculated molecular mass of approximately 13 kDa. Recombinant Interleukin-15 is a disulfide-linked homodimeric protein consisting of two 115 amino acid residue subunits, and migrates as an approximately 13kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.

L15 has been shown to be produced by human fetal astrocytes and microglial cells in response to IL1- beta, IFN-gamma, or TNF- alpha and may thus play a role in T-cell mediated immune responses in the human central nervous system. IL15 stimulates proliferation of the established T-cell line CTLL-2, CD8 (+) memory T- cells require IL15 for proliferation.IL15 is also able to induce generation of cytolytic cells and LAK cells activity in vitro. IL15 appears to function as a specific maturation factor for NK-cells and can mimic the bone marrow microenvironment in vitro, which is required for NK- cells to acquire lytic functions. L15 induces proliferation of mast cells in the absence of functional IL2 receptor components and utilizes a receptor of 60-65 kDa that is distinct from known components of T-cell IL15 receptors. The IL15 receptor on mast cells recruits JAK2 and STAT5, instead of JAK1, JAK3 and STAT3 and STAT5 that are activated in T- cells. The alpha subunit of the IL15 receptor (IL15RA) is structurally related to the alpha subunit of the IL2 receptor and confers high affinity binding of IL15 to its receptor. IL15 binds to the beta and gamma chains of the IL2 receptor, which are known to be required for ligand internalization and signal transduction.

NCBI Description

The protein encoded by this gene is a cytokine that regulates T and natural killer cell activation and proliferation. This cytokine and interleukine 2 share many biological activities. They are found to bind common hematopoietin receptor subunits, and may compete for the same receptor, and thus negatively regulate each other's activity. The number of CD8+ memory cells is shown to be controlled by a balance between this cytokine and IL2. This cytokine induces the activation of JAK kinases, as well as the phosphorylation and activation of transcription activators STAT3, STAT5, and STAT6. Studies of the mouse counterpart suggested that this cytokine may increase the expression of apoptosis inhibitor BCL2L1/BCL-x(L), possibly through the transcription activation activity of STAT6, and thus prevent apoptosis. Alternatively spliced transcript variants of this gene have been reported. [provided by RefSeq, Feb 2011]
