Preparation and Storage
For all formats, store at + 4 degree C. For long term storage, aliquot and freeze unused portion at -20 degree C in volumes appropriate for single usage. Avoid freeze/thaw cycles.

Synaptotagmin-1 is a synaptic vesicle membrane protein that belongs to the Synaptotagmins family; characterized by an N-terminal transmembrane region (TMR), a variable linker, and two C-terminal C2-domains. Synaptotagmin-1, the first synaptotagmin studied, is an abundant synaptic vesicle protein that binds Ca2+ and phospholipids via both of its C2 domains and is essential for fast Ca2+- triggered neurotransmitter release. Multiple members (15) exist within the Synaptotagmins family with high homology in their C-terminal repeats and lowest in their N-terminal intravesicular sequences. The two internal repeats in synaptotagmin are homologous to the regulatory region of protein kinase C. Synaptotagmin -1 and 2 are highly homologous but differentially distributed, with Synaptotagmin-1 being highly expressed in the forebrain and some endocrine tissues and Synaptotagmin-2 in the brain stem and spinal cord.

NCBI Description

The synaptotagmins are integral membrane proteins of synaptic vesicles thought to serve as Ca(2+) sensors in the process of vesicular trafficking and exocytosis. Calcium binding to synaptotagmin-1 participates in triggering neurotransmitter release at the synapse (Fernandez-Chacon et al., 2001 [PubMed 11242035]).[supplied by OMIM, Jul 2010]

Function: May have a regulatory role in the membrane interactions during trafficking of synaptic vesicles at the active zone of the synapse. It binds acidic phospholipids with a specificity that requires the presence of both an acidic head group and a diacyl backbone. A Ca2+-dependent interaction between synaptotagmin and putative receptors for activated protein kinase C has also been reported. It can bind to at least three additional proteins in a Ca2+-independent manner; these are neurexins, syntaxin and AP2.

Cofactor: Binds 3 calcium ions per subunit. The ions are bound to the C2 domains
