Rats are immunised with synthetic oligopeptide (containing an N-terminal cysteine)-carrier protein conjugates. IL-10 was conjugated to JES3. The mouse non-secreting myeloma fusion partner P3X63-AG8.653, is fused to immunised rat splenocytes to produce heterohybridomas secreting rat antibody. Bioassays were used to identify neutralising mAbs, and immunochemical screens were used to identify mAbs that bind the cytokine.

The protein encoded by this gene is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Mutations in this gene are associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis.[provided by RefSeq, May 2011]

AAA Biotech's IL-10 can be used in a range of immunoassay formats including, but not limited to, Neutralisation, ELISA (EIA), Immunohistochemistry (IHC), Immunofluorescence (IF), Immunoprecipitation (IP), Crystallization. 9D7 has been used in ELISA experiments in order to specifically detect IL-10 and show higher circulating levels of IL-10 in patients with acute EBV-induced mononucleosis (Taga 1995). Further, 9D7 has been used to immunoprecipitate IL-10 from the supernatants of melanoma cells in order to study the production of IL-10 by melanoma cells (Qiyuan Chen 1994). A fab fragment derived from 9D7 has been used to study the noncompetitive antibody neutralisation of IL-10 by x-ray crystallography (Josephson 2002). Researchers should empirically determine the suitability of the IL-10 il10 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
