Subunit Structure
Component of the I-kappa-B-kinase (IKK) core complex consisting of CHUK, IKBKB and IKBKG; probably four alpha/CHUK-beta/IKBKB dimers are associated with four gamma/IKBKG subunits. The IKK core complex seems to associate with regulatory or adapter proteins to form a IKK-signalosome holo-complex. The IKK complex associates with TERF2IP/RAP1, leading to promote IKK-mediated phosphorylation of RELA/p65. Part of a complex composed of NCOA2, NCOA3, CHUK/IKKA, IKBKB, IKBKG and CREBBP. Part of a 70-90 kDa complex at least consisting of CHUK/IKKA, IKBKB, NFKBIA, RELA, IKBKAP and MAP3K14. Directly interacts with IKK-gamma/NEMO and TRPC4AP. May interact with TRAF2. Interacts with NALP2. May interact with MAVS/IPS1. Interacts with ARRB1 and ARRB2. Interacts with NLRC5; prevents CHUK phosphorylation and kinase activity. Interacts with PIAS1; this interaction induces PIAS1 phosphorylation. Interacts with ZNF268 isoform 2; the interaction is further increased in a TNF-alpha-dependent manner. / Component of the I-kappa-B-kinase (IKK) core complex consisting of CHUK, IKBKB and IKBKG; probably four alpha/CHUK-beta/IKBKB dimers are associated with four gamma/IKBKG subunits. Entrez Summary: This gene encodes a member of the serine/threonine protein kinase family. The encoded protein, a component of a cytokine-activated protein complex that is an inhibitor of the essential transcription factor NF-kappa-B complex, phosphorylates sites that trigger the degradation of the inhibitor via the ubiquination pathway, thereby activating the transcription factor. [provided by RefSeq, Jul 2008]/The protein encoded by this gene phosphorylates the inhibitor in the inhibitor/NF-kappa-B complex, causing dissociation of the inhibitor and activation of NF-kappa-B. The encoded protein itself is found in a complex of proteins. Several transcript variants, some protein-coding and some not, have been found for this gene. [provided by RefSeq, Sep 2011]

Uniprot Description

IKKA: a kinase of the IKK family. Phosphorylates inhibitors of NF-kappa-B, leading to their dissociation from NF-kappa-B and ultimately to their degradation. Phosphorylated and activated downstream of growth factor receptors, IL1R, and TNFR by NAK, IRAK and NIK, respectively. Preferentially found as a heterodimer with IKK-beta but also as an homodimer. Directly interacts with IKK-gamma/NEMO and TRPC4AP. Heterodimers form the active complex. The tripartite complex can also bind to MAP3K14/NIK, MEKK1, IKAP and IKB-alpha-p65-p50 complex. Inhibitors are under development to treat arthritis, inflammation, and apoptotic aspects of cancer. Aspirin (sodium salicylate) selectively binds IKK and reduces inflammation. Misexpression and inhibitors show involvement in insulin sensitization in obese rodents. Mutations in IKK-gamma are found in several immune deficiencies: hyper IgM syndrome, incontinentia pigmenti [embryonic lethal in males, defects in skin, hair, teeth in females] and hypohidrotic ectodermal dysplasia; recurrent infections and defects in teeth, hair, and sweat glands]. Inhibitors: CHS 828, NBD peptide, BMS-345541.

Product Notes

The IKKa/b chuk (Catalog #AAA477146) is an Antibody produced from Rabbit Source: Human and is intended for research purposes only. The product is available for immediate purchase. The Anti-IKKa/b (Phospho-Ser180/181) Rabbit pAb reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's IKKa/b can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC). Researchers should empirically determine the suitability of the IKKa/b chuk for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IKKa/b, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
