NCBI Description

The protein encoded by this gene is a major component of the postsynaptic density (PSD), a group of proteins found associated with NMDA receptors at synapses. The encoded protein is phosphorylated by calmodulin-dependent protein kinase II and dephosphorylated by NMDA receptor activation. Defects in this gene are a cause of mental retardation autosomal dominant type 5 (MRD5). [provided by RefSeq, Dec 2009]

Uniprot Description

SynGAP: an excitatory synapse-specific RasGAP. Member of the NMDAR signaling complex in excitatory synapses it may play a role in NMDAR-dependent control of AMPAR potentiation and synaptic plasticity. ERK activation is up-regulated in neurons from SynGAP knockout mice, whereas P38 MAPK function is depressed. May play a critical role in the regulation of neuronal MAPK signaling, AMPAR membrane trafficking, and excitatory synaptic transmission. Interacts with CAMK2A/CAMK2B and MPDZ. Phosphorylated by CaM-kinase II. Dephosphorylated upon NMDA receptor activation or SYNGAP1/MPDZ complex disruption. Contains a PH domain and a Ras-GAP domain. Two alternatively spliced isoforms have been described.

Protein type: GAPs, Ras; GAPs

Chromosomal Location of Human Ortholog: 6p21.3

Cellular Component: cytoplasm; cytosol; dendritic shaft

Molecular Function: GTPase activator activity; protein kinase binding; SH3 domain binding

Biological Process: dendrite development; MAPKKK cascade; negative regulation of axonogenesis; negative regulation of neuron apoptosis; negative regulation of Ras protein signal transduction; pattern specification process; positive regulation of GTPase activity; Ras protein signal transduction; receptor clustering; regulation of long-term neuronal synaptic plasticity; regulation of MAPKKK cascade; regulation of synaptic plasticity; visual learning

Product Notes

The SYNGAP1 syngap1 (Catalog #AAA422696) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Goat anti-SYNGAP1 (aa60-72) Antibody reacts with Tested: Mouse; Expected from sequence similarity: Human, Mouse, Rat, Dog, Pig, Cow and may cross-react with other species as described in the data sheet. AAA Biotech's SYNGAP1 can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA), Western Blot (WB). Peptide ELISA: Antibody detection limit dilution 1: 32000. Western Blot: Approx 150kDa band observed in Mouse Brain lysates (calculated MW of 148kDa according to Human NP_006763.2 and Mouse XP_990642.3). Recommended concentration: 1-3ug/ml. Researchers should empirically determine the suitability of the SYNGAP1 syngap1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: DEDEIHPLLI RDR. It is sometimes possible for the material contained within the vial of "SYNGAP1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
