Description: This assay employs a two-site sandwich ELISA to quantitate MOSPD3 in samples. An antibody specific for MOSPD3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MOSPD3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MOSPD3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MOSPD3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Pall et al. (2004) cloned mouse Mospd3 and identified the human MOSPD3 gene. The deduced 235-amino acid MOSPD3 protein contains an N-terminal major sperm protein domain and 2 transmembrane domains. By genomic sequence analysis, Pall et al. (2004) mapped the MOSPD3 gene to chromosome 7q22. They mapped the mouse Mospd3 gene to a region of chromosome 5 that shows homology of synteny to human chromosome 7q22. Pall et al. (2004) found that homozygous Mospd3-null mice had a right ventricle defect with 50% probability of neonatal lethality. The phenotypic defect was dependent on the genetic background, indicating the presence of modifier loci

This gene encodes a multi-pass membrane protein with a major sperm protein (MSP) domain. The deletion of a similar mouse gene is associated with defective cardiac development and neonatal lethality.
