Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate RTTN in samples. An antibody specific for RTTN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyRTTN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RTTN is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RTTN bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a large protein whose specific function is unknown. Absence of the orthologous protein in mouse results in embryonic lethality with deficient axial rotation, abnormal differentiation of the neural tube, and randomized looping of the heart tube during development. In human, mutations in this gene are associated with polymicrogyria with seizures. In human fibroblasts this protein localizes at the ciliary basal bodies. Given the intracellular localization of this protein and the phenotypic effects of mutations, this gene is suspected of playing a role in the maintenance of normal ciliary structure which in turn effects the developmental process of left-right organ specification, axial rotation, and perhaps notochord development.
