Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate MYL2 in samples. An antibody specific for MYL2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMYL2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MYL2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MYL2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Contractile protein that plays a role in heart development and function (). Following phosphorylation, plays a role in cross-bridge cycling kinetics and cardiac muscle contraction by increasing myosin lever arm stiffness and promoting myosin head diffusion; as a consequence of the increase in maximum contraction force and calcium sensitivity of contraction force. These events altogether slow down myosin kinetics and prolong duty cycle resulting in accumulated myosins being cooperatively recruited to actin binding sites to sustain thin filament activation as a means to fine-tune myofilament calcium sensitivity to force (). During cardiogenesis plays an early role in cardiac contractility by promoting cardiac myofibril assembly ().
