EEF1d encodes a subunit of the elongation factor-1 complex, which is responsible for the enzymatic delivery of aminoacyl tRNAs to the ribosome. This subunit functions as guanine nucleotide exchange factor. It is reported that this subunit interacts with HIV-1 Tat, and thus it represses the translation of host-cell, but not HIV-1, mRNAs. Several alternatively spliced transcript variants have been found for this gene, however, the full length nature of only two variants has been determined. The deduced 281-amino acid protein has a calculated molecular mass of 31 kD. It has an N-terminal leucine zipper domain and a C-terminal domain that is similar to that of EF1-beta and is predicted to show GDP/GTP exchange activity. EF1-delta also has a conserved serine phosphorylation site.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate EEF1D in samples. An antibody specific for EEF1D has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyEEF1D present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EEF1D is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EEF1D bound in the initial step. The color development is stopped and the intensity of the color is measured.
