Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate TNNT2 in samples. An antibody specific for TNNT2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNNT2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNNT2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNNT2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

TNNT2: Troponin T is the tropomyosin-binding subunit of troponin, the thin filament regulatory complex which confers calcium-sensitivity to striated muscle actomyosin ATPase activity. Heart. The fetal heart shows a greater expression in the atrium than in the ventricle, while the adult heart shows a greater expression in the ventricle than in the atrium. Isoform 6 predominates in normal adult heart. Isoforms 1, 7 and 8 are expressed in fetal heart. Isoform 7 is also expressed in failing adult heart. Belongs to the troponin T family. 11 isoforms of the human protein are produced by alternative splicing.
