Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate HARS2 in samples. An antibody specific for HARS2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyHARS2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HARS2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HARS2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

Aminoacyl-tRNA synthetases are a class of enzymes that charge tRNAs with their cognate amino acids. The protein encoded by this gene is an enzyme belonging to the class II family of aminoacyl-tRNA synthetases. Functioning in the synthesis of histidyl-transfer RNA, the enzyme plays an accessory role in the regulation of protein biosynthesis. The gene is located in a head-to-head orientation with HARS on chromosome five, where the homologous genes likely share a bidirectional promoter. Mutations in this gene are associated with the pathogenesis of Perrault syndrome, which involves ovarian dysgenesis and sensorineural hearing loss.
