Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate NMT1 in samples. An antibody specific for NMT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNMT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NMT1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NMT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

Myristate, a rare 14-carbon saturated fatty acid, is cotranslationally attached by an amide linkage to the N-terminal glycine residue of cellular and viral proteins with diverse functions. N-myristoyltransferase (NMT; EC 2.3.1.97) catalyzes the transfer of myristate from CoA to proteins. N-myristoylation appears to be irreversible and is required for full expression of the biologic activities of several N-myristoylated proteins, including the alpha subunit of the signal-transducing guanine nucleotide-binding protein (G protein) GO (GNAO1; MIM 139311) (Duronio et al., 1992 [PubMed 1570339]).[supplied by OMIM, Nov 2008]
