Intended Uses: For the quantitative detection of Human Phosphoenolpyruvate carboxykinase [GTP], mitochondrial (PCK2) concentration in serum, plasma, tissue homogenates and other biological fluids.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate PCK2 in Human serum, plasma, tissue homogenates. An antibody specific for PCK2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PCK2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PCK2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PCK2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a mitochondrial enzyme that catalyzes the conversion of oxaloacetate to phosphoenolpyruvate in the presence of guanosine triphosphate (GTP). A cytosolic form of this protein is encoded by a different gene and is the key enzyme of gluconeogenesis in the liver. Alternatively spliced transcript variants have been described.
