Application Notes
1. To quickly determine which Sample Diluent formulation is best for the assay, all three Sample Diluents can be run in parallel on the same IgG or antigen-coated ELISA plate.
2. Determine the initial dynamic analyte range of the ELISA curves that were prepared in each of the three different Sample Diluent types.
3. Make test spikes into serum samples.
4. Read the spiked serum sample wells on each of the three SD-associated standard curves to see which Sample Diluent gives the best approximation of the spike concentration that was added to the serum samples.
5. In cases where a high level of matrix complexity interference is detected, inclusion of an additional Assay Diluent reagent may also be required to bring the calibrator well antibody-to-antigen binding kinetics closer to the sample well binding kinetics.

Description: Sample Diluent Optimization Pack provides three 100 mL bottles of Sample Diluent formulations (General Serum Diluent, Plasma Sample Diluent, and Neptune Sample Diluent) for an economical and fast method of selecting the best Sample Diluent for a particular assay.

Background: When developing any new ELISA, it is often helpful to compare a number of different sample diluent formulations to better match the matrix complexity present in the sample wells with the standard curve calibrator matrix wells. Most of the time invested in developing a new ELISA/EIA/RIA will involve selecting the proper calibrator matrix that matches most closely to the often very complex biological sample matrix that is being evaluated. Comparing three different sample/calibrator diluent formulations on the same IgG or antigen-coated ELISA plate makes it is easy to determine which diluent will best equalize the calibrator well antibody-binding kinetics with the biological sample/ dilution well antibody-binding kinetics. The Sample Diluent Optimization Pack provides 100 mL of each of our three Sample Diluent formulations to begin optimizing the sample diluent portion of the assay development process.

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Sample Diluent Optimization Pack, Diluent" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
