Preparation and Storage
Storage:
Avoid repeated freeze/thaw cycles.
Store at 4ºC for frequent use.
Aliquot and store at -20ºC for 24 months.

Stability Test:
The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37°C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

NCBI Description

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. This structure consists of approximately 146 bp of DNA wrapped around a nucleosome, an octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a replication-dependent histone that is a member of the histone H2A family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6p22-p21.3. Uniprot Description

HIST1H2AD: Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. Belongs to the histone H2A family. The nucleosome is a histone octamer containing two molecules each of H2A, H2B, H3 and H4 assembled in one H3-H4 heterotetramer and two H2A-H2B heterodimers. The octamer wraps approximately 147 bp of DNA

Chromosomal Location of Human Ortholog: 6p22.2

Cellular Component: nuclear chromatin; nucleosome; nucleus

Molecular Function: DNA binding; protein heterodimerization activity

Biological Process: chromatin silencing

Product Notes

The HIST1H2AD hist1h2ad (Catalog #AAA2005055) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Polyclonal Antibody to Histone Cluster 1, H2ad (HIST1H2AD) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Histone Cluster 1, H2ad (HIST1H2AD) can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), Immunocytochemistry (ICC), Immunoprecipitation (IP). Western blotting: 0.5-2ug/mL Immunohistochemistry: 5-20ug/mL Immunocytochemistry: 5-20ug/mL Optimal working dilutions must be determined by end user. Researchers should empirically determine the suitability of the HIST1H2AD hist1h2ad for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Histone Cluster 1, H2ad (HIST1H2AD), Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
