Preparation and Storage
For unopened kit, all the reagents should be kept according to the label on each vial. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C while others should be at 4 degree C.

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to WW Domain Containing Transcription Regulator Protein 1 (WWTR1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to WW Domain Containing Transcription Regulator Protein 1 (WWTR1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain WW Domain Containing Transcription Regulator Protein 1 (WWTR1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of WW Domain Containing Transcription Regulator Protein 1 (WWTR1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

TAZ: Transcriptional coactivator which acts as a downstream regulatory target in the Hippo signaling pathway that plays a pivotal role in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. The core of this pathway is composed of a kinase cascade wherein STK3/MST2 and STK4/MST1, in complex with its regulatory protein SAV1, phosphorylates and activates LATS1/2 in complex with its regulatory protein MOB1, which in turn phosphorylates and inactivates YAP1 oncoprotein and WWTR1/TAZ. WWTR1 enhances PAX8 and NKX2-1/TTF1-dependent gene activation. Regulates the nuclear accumulation of SMADS and has a key role in coupling them to the transcriptional machinery such as the mediator complex. Regulates embryonic stem-cell self-renewal, promotes cell proliferation and epithelial-mesenchymal transition. Binds to SLC9A3R2 via the PDZ motif at the plasma membrane. Binds to YWHAZ in vivo and in vitro through the phosphoserine-binding motif RSHSSP. Interacts (via coiled-coil domain) with SMAD2 (via MH1 domain), SMAD3 and SMAD4. Interacts with MED15, PAX8 and NKX2-1. Interacts with TEAD1, TEAD2, TEAD3 and TEAD4. Highly expressed in kidney, heart, placenta and lung. Expressed in the thyroid tissue.
