Principle of the Assay: The Picokine™ Human CCL19 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human CCL19 with a 96-well strip plate that is pre-coated with antibody specific for CCL19. The detection antibody is a biotinylated antibody specific for CCL19. The capture antibody is monoclonal antibody from mouse, the detection antibody is polyclonal antibody from goat. The kit contains recombinant Human CCL19 with immunogen: Expression system for standard: E.coli; Immunogen sequence: G22-S98. The kit is analytically validated with ready to use reagents. To measure Human CCL19, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human CCL19 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human CCL19 in the sample.!!Background/Introduction: Chemokine(C-C motif) ligand 19(CCL19) is a small cytokine belonging to the CC chemokine family that is also known as EBI1 ligand chemokine(ELC) and macrophage inflammatory protein-3-beta (MIP-3-beta). Using PCR of human-rodent hybrids, the CCL19 gene was map to human chromosome 9p13, and the CCL19 gene is not part of the CC chemokine gene cluster on chromosome 17. The cytokine encoded by this gene may play a role in normal lymphocyte recirculation and homing. It also plays an important role in trafficking of T cells in thymus, and in T cell and B cell migration to secondary lymphoid organs.

NCBI Description

This antimicrobial gene is one of several CC cytokine genes clustered on the p-arm of chromosome 9. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene may play a role in normal lymphocyte recirculation and homing. It also plays an important role in trafficking of T cells in thymus, and in T cell and B cell migration to secondary lymphoid organs. It specifically binds to chemokine receptor CCR7.
