Introduction: Mediator of ErbB2-driven cell motility 1 (MEMO1) is a member of the UPF0103 family. MEMO1 regulates cell migration by transmitting extracellular chemotactic signals to the microtubule cytoskeleton. Furthermore, MEMO1 controls the localization of APC and CLASP2 to the cell membrane, using the regulation of GSK3B activity. MEMO1 is essential for breast carcinoma cell migration, suggesting a key role in tumorigenesis. MEMO1 is also a mediator of ERBB2 signaling.

MEMO1: a highly conserved protein that controls cell migration by relaying extracellular chemotactic signals to the microtubule cytoskeleton. Regulates directionality during HER2-dependent cell migration. Controls microtubules, the actin network, and adhesion site formation in migrating cells via the MEMO1-RHOA-DIAPH1 signaling pathway. Homologous to class III nonheme iron-dependent dioxygenases. No metal-binding or enzymatic activity has been detected. Binds HER2 phosphorylated at Y1222 via its vestigial enzymatic active site. Binds to and influences insulin-like growth factor-I (IRS1) receptor-dependent signaling events in breast carcinogenesis. Influences the epithelial-mesenchymal transition (EMT) via the IRS1/PI3K/Akt/Snail1 pathway. Interacts with cofilin, and influences PLCg1 and cofilin activities. Increased expression is associated with cancer aggressiveness. The MEMO1-RHOA-DIAPH1 signaling pathway plays an important role in ERBB2-dependent stabilization of microtubules at the cell cortex. It controls the localization of APC and CLASP2 to the cell membrane, via the regulation of GSK3B activity. In turn, membrane-bound APC allows the localization of the MACF1 to the cell membrane, which is required for microtubule capture and stabilization. Two isoform of the human protein are produced by alternatuve splicing.
