Application Notes
AURKB antibody has been tested by ELISA and Immunofluorescence analyses to assure specificity and reactivity. Since applications vary, each investigation should be titrated by a reagent to obtain optimal results. Recommended dilution range for Immunofluorescence is 1:1000 ~ 2000. Recommended starting dilution is 1:1000.
Immunogen
Anti-human AURKB mAb, is derived from hybridization of mouse F0 myeloma cells with spleen cells from BALB/c mice immunized with recombinant human AURKB amino acids 1-344 purified from E Coli.
Preparation and Storage
For periods up to 1 month store at 4 degree C, for longer periods of time, store at -20 degree C. Prevent freeze thaw cycles.

Introduction: Aurora Kinase B (AURKB) belongs to a family of mitotic serine/threonine kinases. AURKB connects with chromosomes for the period of prophase prior to relocalizing to the spindle at anaphase. AURKB localizes to microtubules near kinetochores, specifically to the specialized microtubules called K-fibers. AURKB controls chromosome segregation through the control of microtubule-kinetochore attachment and cytokinesis. AURKB is required for kinetochore localization of BUB1 and SGOL1. AURKB expression during the G2/M phase transition is firmly coordinated with histone H3 phosphorylation, while overexpression is seen in many kinds of cancers. AURKB phosphorylates ''Ser-10'' and ''Ser-28'' of histone H3 during mitosis. AURKB is a component of the CPC (chromosomal passenger complex), which is a complex that acts as a key regulator of mitosis. High level expression of AURKB is seen in the thymus, which is also expressed in the spleen, lung, testis, colon, placenta and fetal liver. AURKB is expressed during S and G2/M phase and expression is up-regulated in cancer cells during M phase.

NCBI Description

This gene encodes a member of the aurora kinase subfamily of serine/threonine kinases. The genes encoding the other two members of this subfamily are located on chromosomes 19 and 20. These kinases participate in the regulation of segregation of chromosomes during mitosis and meiosis through association with microtubules. A pseudogene of this gene is located on chromosome 8. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jan 2013]

AurB: a member of the AUR family of kinases. May be directly involved in regulating the cleavage of polar spindle microtubules and is a key regulator for the onset of cytokinesis during mitosis. Expressed during S and G2/M phase and expression is upregulated in cancer cells during M phase. Localized to the midzone of central spindle in late anaphase and concentrated into the midbody in telophase and cytokinesis. Colocalizes with gamma tubulin in the mid-body. Component of the chromosomal passenger complex (CPC), a complex that acts as a key regulator of mitosis. The CPC complex has essential functions at the centromere in ensuring correct chromosome alignment and segregation and is required for chromatin-induced microtubule stabilization and spindle assembly. Overexpressed in colorectal and other cancer cell lines and thought to cause aneuploidy via histone phosphorylation.
