Application Notes
HOMER1 antibody has been tested by ELISA and Western blot analysis to assure specificity and reactivity. Since application varies, however, each investigation should be titrated by the reagent to obtain optimal results. Recommended dilution range for Western blot analysis is 1:1000 ~ 2000. Recommended starting dilution is 1:1000.
Immunogen
Anti-human HOMER1 mAb is derived from hybridization of mouse F0 myeloma cells with spleen cells from BALB/c mice immunized with recombinant human HOMER1 amino acids 1-354 purified from E Coli.
Preparation and Storage
For periods up to 1 month store at 4 degree C, for longer periods of time, store at -20 degree C. Prevent freeze thaw cycles.
12 months at -20 degree C. 1 month at 4 degree C.

Introduction: HOMER1 takes part in determining the apoptotic susceptibility to TRAIL. HOMER1 is a member of the Homer family of proteins that regulate signal transduction through, and the trafficking of, glutamate receptors, plus maintain and regulate extracellular glutamate levels in corticolimbic brain regions. The HOMER1 is enriched at excitatory synapses and binds group 1 metabotropic glutamate receptors (mGluRs).

NCBI Description

This gene encodes a member of the homer family of dendritic proteins. Members of this family regulate group 1 metabotrophic glutamate receptor function. [provided by RefSeq, Jul 2008]

Uniprot Description

HOMER1: Postsynaptic density scaffolding protein. Binds and cross-links cytoplasmic regions of GRM1, GRM5, ITPR1, DNM3, RYR1, RYR2, SHANK1 and SHANK3. By physically linking GRM1 and GRM5 with ER-associated ITPR1 receptors, it aids the coupling of surface receptors to intracellular calcium release. May also couple GRM1 to PI3 kinase through its interaction with AGAP2. Isoform 1 regulates the trafficking and surface expression of GRM5. Isoform 3 acts as a natural dominant negative, in dynamic competition with constitutively expressed isoform 1 to regulate synaptic metabotropic glutamate function. Isoform 3, may be involved in the structural changes that occur at synapses during long-lasting neuronal plasticity and development. Interacts with IFT57. Interacts with OPHN1. Isoform 1 encodes a coiled-coil structure that mediates homo- and heteromultimerization. Belongs to the Homer family. 3 isoforms of the human protein are produced by alternative splicing.
